Not sure where it was made, but I know it was sent from Canada. and am very pleased with it. The price that it was advertised at was what I paid as I live in the U.K nothing to pay after it got into the country as it included the taxes .

What size line can you use with this?

1/2 inch docking line works great...

What is the nylon screw for?

It holds the line in place. You back the screw out a bit to slip the line into the boat hook, tie your bowline, then tighten the screw down a bit to hold the base of the bowline up against the stick. Otherwise, it'd be harder to keep the loop open.
